What Is a Certificate of Analysis (COA) and Why Researchers Need One
In research peptide sourcing, the Certificate of Analysis (COA) is the single most important document a supplier can provide. It is the difference between knowing what you are working with and trusting a label. For any researcher working with peptides, understanding what a COA contains and what to look for is not optional.
What a COA Is
A Certificate of Analysis is a document produced by an analytical laboratory that has tested a specific lot of a compound. It reports the results of testing performed to verify the identity, purity, and quality of the compound. A legitimate COA is produced by an independent third-party laboratory, not by the supplier itself. Internal QC documents produced by the supplier are not COAs in the meaningful sense.
What a Good COA Contains
A complete COA for a research peptide should include several key elements. First, compound identity confirmation, typically through mass spectrometry (MS) or high-performance liquid chromatography (HPLC) data showing that the molecular weight and structure match the expected compound. Second, purity percentage, usually expressed as a percentage of the total area under the HPLC curve attributable to the target peptide. Third, lot or batch number, allowing the COA to be matched to a specific production run. Fourth, date of testing. Fifth, the name and ideally accreditation details of the laboratory that performed the testing.
How to Read Purity Data
HPLC purity is the most commonly reported purity metric for peptides. A purity of 98% or above is generally considered high-quality for research purposes. Some suppliers report 99% or greater. Purity below 95% should be viewed with caution for research applications because the impurities are undefined and could affect results.
The HPLC chromatogram itself (if included in the COA or available on request) shows the peaks corresponding to the target peptide and any impurities. Reviewing the chromatogram directly provides more information than the summary purity percentage alone.
Mass Spectrometry: Confirming Identity
Purity alone is not enough: a sample could be pure but consist of a different peptide entirely. Mass spectrometry data confirms the molecular weight of the compound, which is specific to each peptide sequence. The observed mass should match the theoretical mass for the compound within the acceptable tolerance of the instrument used.
A COA that includes both HPLC purity and MS identity data provides much stronger assurance than one that provides only purity or only identity.
Red Flags in COA Documentation
Several patterns indicate COA documentation that should not be trusted. COAs that are clearly generic (no batch number, no date, no laboratory name) are not meaningful. COAs dated years ago for current stock are suspicious: fresh lots should have recent testing. COAs from laboratories that cannot be independently verified are a concern. Suppliers who cannot provide COAs on request or who provide them only after purchase are not operating transparently.
Endotoxin Testing
For some research applications, endotoxin (lipopolysaccharide) contamination testing is also important. Bacterial endotoxins can cause significant inflammatory responses and confound research results. Not all COAs include endotoxin testing, but for applications where this would affect results, it is worth requesting specifically.

Research conducted with an impure or misidentified compound produces invalid results. The time, cost, and validity of research depends on compound integrity.
