What Is Retatrutide?

How Does Retatrutide Work?

Glucagon receptor activation increases energy expenditure, meaning the body burns more calories at rest. Semaglutide (the active compound in Ozempic and Wegovy) targets one receptor: GLP-1. Tirzepatide (the active compound in Mounjaro) targets two: GLP-1 and GIP. Retatrutide targets all three. In the Phase 2 clinical trial published in the New England Journal of Medicine in 2023, participants taking the highest dose of retatrutide lost an average of 24% of their body weight over 48 weeks, a figure that exceeded results seen in comparable trials for semaglutide and tirzepatide.

What Does the Research Show?

The 2023 Phase 2 trial was the landmark study. It enrolled 338 adults with obesity and showed dose-dependent weight reduction across all groups, with the highest dose group achieving the 24% average figure. Retatrutide also showed improvements in blood sugar, blood pressure, and lipid levels. Phase 3 trials are ongoing as of 2026.

Retatrutide Side Effects: What Research Has Documented

The most commonly reported side effects in trials were gastrointestinal: nausea, vomiting, diarrhea, and constipation, similar to those seen with semaglutide and tirzepatide. These were generally dose-dependent, meaning higher doses produced more frequent side effects. Most were reported as mild to moderate. The trial also noted a small increase in heart rate, which is being monitored in ongoing Phase 3 studies.
